Efficient cell lysis and maximum protein extraction yields are vital to high-quality recombinant protein purification. The protease inhibitor cocktails are for lysing mammalian cells, buffers that solubilize plasma and intracellular membranes, break weak intermolecular bonds, and solubilize most proteins. Making procedures easier to perform, these protein extraction reagents can also be used to isolate proteins from both the cytoplasmic and nuclear fractions of cultured cells.

Beskrivning: FLUOR DE LYS® Developer is an essential reagent for signal generation in HDAC or sirtuin assays done with the FLUOR DE LYS® substrate. An appropriate FLUOR DE LYS® family substrate, comprising an acetylated lysine side chain, is first incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes it so that, in a second step, treatment with the FLUOR DE LYS® Developer produces a fluorophore. Typically, an ‘inhibitor stop’ will be included in the 1x Developer dilution (e.g. Trichostatin A) for class I and II HDACs, nicotinamide for sirtuins).

A crucial step in using peptides as antigens is their coupling to carrier proteins. Peptides are designed to be good epitopes by themselves; however they fail to elicit a strong antibody response. The reason for this is that the epitopes are able to bind to the B-cells, but are too small to contain additional motifs required to associate with class II molecules and T-cell receptors. The carrier proteins, once coupled to the peptides, provide these additional motifs and generate a strong antibody response. All three carrier proteins are also offered as activated carrier proteins, named as ActiveHOOK™.
