Beskrivning: 3,3′-Diaminobenzidine has been used as a chromogen for the visualisation in immunohistochemistry of chondrosarcoma sections, mammary carcinomas and brain tissue sections. Peroxidase substrate; reagent for spectrophotometric determination of selenium. DAB (3,3′-Diaminobenzidine) is utilized in many applications for the visualisation of peroxidase activity. In the peroxidase reaction, DAB serves as a hydrogen donor in the presence of peroxide. The oxidised DAB forms an insoluble brown end-product for use in the immunohistological and immunoblotting staining procedures.

Selenium exhibits potent anticarcinogenic properties, and deficiency of selenium may cause certain neurologic diseases. Both effects are attributed to selenium-binding proteins. Selenium binding protein 1 is down-regulated in lung adenocarcinoma, colorectal cander and ovarian cancer. It is two-fold upregulated in the brains of patients suffering from schizophrenia, and is therefore a biomarker for this disease.

Iscove's medium is a modification of Dulbecco's Modified Eagle's Medium (DME) containing selenium, additional amino acids and vitamins, sodium pyruvate, HEPES buffer, and potassium nitrate instead of ferric nitrate. Further studies have demonstrated that Iscove's medium will support murine B lymphocytes, haemopoietic tissue from bone marrow, B-cells stimulated with lipopolysaccharide, T-lymphocytes, and a variety of hybrid cells.

There are eight GPx homologs (GPx-1–8). GPx-1 plays an important role in the antioxidant defense of the vascular wall and neural cells in response to oxidative stress. GPx-2 is the major isoform in the lungs and its basal or inducible expression is dependent on Nrf2. GPx-3 is under regulation by hypoxic stress and the expression and deficiency of GPx-3 is associated with cardiovascular disease and stroke. GPx-5 is selenium-independent; it is bound to the acrosome of sperm, where it may protect sperm from premature acrosome reaction in the epididymis.

Beskrivning: Glutathione peroxidases (Gpxs) are ubiquitously expressed proteins which catalyze the reduction of hydrogen peroxides and organic hydroperoxides by glutathione. There are several isoforms which differ in their primary structure and localization. The classical cytosolic/mitochondrial GPx1 (cGPx) is a selenium-dependent enzyme, first of the GPx family to be discovered. GPx2, also known as gastrointestinal GPx (GI-GPx), is an intracellular enzyme expressed only at the epithelium of the gastrointestinal tract. Extracellular plasma GPx (pGPx or GPx3) is mainly expressed by the kidney from where it is released into the blood circulation. Phospholipid hydroperoxide GPx4 (PH-GPx) expressed in most tissues, can reduce many hydroperoxides including hydroperoxides integrated in membranes, hydroperoxy lipids in low density lipoprotein or thymine. All mammalian GPx family members, except for the recently described Cys containing GPx3 and epididymis-specific secretory GPx (eGPx or GPx5) isoforms, possess selenocysteine at the active site.
