Beskrivning: Melanocytes produce organelles called melanosomes which produce melanin, the pigment that gives color to skin, hair, eyes, scales and feathers. gp100 was identified in an attempt to clone the gene Tyrosinase, an enzyme required for melanin synthesis. Further testing determined that gp100 is a melanoma-specific protein and is responsible for melanosome maturation, facilitating the transition from amorphous rounded vesicles to fibrillary ellipsoid organelles.
Metastatic amelanotic melanoma can often be confused with a variety of poorly differentiated carcinomas, large cell lymphomas, and sarcomas using H & E stains alone. It is also difficult to differentiate melanoma from spindle cell carcinomas and various types of mesenchymal neoplasms. Clone HMB45 gp100 antibody stains fetal and neonatal melanocytes, junctional and blue nevus cells, and malignant melanoma. It also stains angiomyolipomas, tumors most commonly associated with the kidney. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. This gp100 antibody does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.

Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. AZGP1 has been reported to stimulate lipid breakdown and may have an important role in lipid homeostasis. Mature human AZGP1 consists of one MHC class I antigen region and a C2-type Ig-like domain. Beskrivning: The HMB45 clone reacts with a neuraminidase-sensitive oligosaccharide side chain of a glycoconjugate present in immature melanosomes. The HMB45-reactive antigen is present in cutaneous melanocytes, prenatal and infantile retinal pigment epithelium and melanoma cells and is thought to be oncofetal in nature. This antibody has been shown to label the majority of melanomas. Clone A103 recognizes a protein of 20 kDa, identified as MART-1 (melanoma antigen recognized by T cells-1) or Melan-A. Melan-A is a useful addition to melanoma panels as it is apparently specific for melanocytic lesions. Studies have also shown that MART-1 is more sensitive than HMB45 when labeling metastatic melanomas. Tyrosinase is a key enzyme involved in the initial stages of melanin biosynthesis. Studies have shown tyrosinase to be a more sensitive marker when compared to HMB45 and MART-1. It has been shown to label a higher percentage of desmoplastic melanomas than HMB45. The combination of HMB45, MART-1 and tyrosinase make this triple antibody cocktail a first-order pan melanoma screener.
