Beskrivning: The 2H7 antibody reacts with human CD20, expressed by pre-B cells, resting and activated B cells, follicular dendritic cells and a small subset of T cells, but not on plasma cells. CD20 is a 33-37 kDa transmembrane protein which is differentially phosphorylated in resting and active B cells. The CD20 molecule plays a role in regulating B-cell activation. Beskrivning: Recognizes a protein of 12 kDa, identified as β-2 microglobulin. Major histocompatibility complex (MHC) class 1 molecules bind to antigens for presentation on the surface of cells. The proteasome is responsible for producing these antigens from the components of foreign pathogens. MHC class 1 molecules consist of an α heavy chain that contains three subdomains (α1, α2, α3) and a non-covalent associating light chain, known as β-2-Microglobulin. β-2-Microglobulin associates with the α3 subdomain of the α heavy chain and forms an immunoglobulin domain-like structure that mediates proper folding and expression of MHC class 1 molecules. The α1 and α2 domains of the α heavy chain form the peptide antigen-binding cleft. Mutations in the β-2-Microglobulin gene can enhance the progression of malignant melanoma phenotypes.

Beskrivning: This MAb recognizes a protein of 145 kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

Beskrivning: Recognizes a protein of 12 kDa, identified as β-microglobulin. β-microglobulin non-covalently associates with the 44 kDa chain to form the HLA Class I antigen complex. Human β-2 microglobulin associated with HLA Class I antigens is expressed on many types of cells including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial cells, and epithelial cells. It is absent on erythrocytes. This MAb is specific to human β-2 microglobulin and does not react with non-human primate cells. This antibody reacts with all cell types excluding erythrocytes. The detection of β-2 microglobulin in body fluids has been used as a tumor marker and for monitoring patients with HIV infection.

Beskrivning: β2 microglobulin is a 12 kDa protein with a pI of 5.6. Serum β2 microglobulin levels are a reflection of cell turnover. Levels rise with fever, inflammation, and infection. Increased serum levels are also seen in B-cell malignancies and in renal failure and may indicate a worse prognosis for patients with early-stage Hodgkin's lymphoma. In urine, increased levels are seen in proximal renal tubular disease as well as renal transplant rejection. β2 microglobulin levels can rise either because its rate of synthesis has increased (e.g. in AIDS, malignant monoclonal plasma cell dyscrasia, solid tumours and autoimmune disease) or because of impaired renal filtration (e.g. due to renal insufficiency, graft rejection or nephrotoxicity induced by post-transplantation immunosuppressive therapy).

Beskrivning: This antibody recognizes an oncofetal glycoprotein with a single chain of 70 kDa, which is identified as alpha fetoprotein (AFP). This MAb is highly specific to AFP and shows no cross-reaction with other oncofetal antigens or serum albumin. The yolk sac and the liver produce AFP during fetal life. AFP expression in adults is often associated with hepatoma or teratoma. However, hereditary persistence of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the AFP and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. AFP is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of AFP in amniotic fluid is used to measure renal loss of protein to screen for spinal bifida and anencephaly.

It is a highly regulated enzyme that catalyzes the hydrolytic deamination of adenosine monophosphate to inosine monophosphate, a branch point in the adenylate catabolic pathway. AMPD3 gene encodes the erythrocyte (E) isoforms, whereas other family members encode isoforms that predominate in muscle (M) and liver (L) cells. This MAb shows reactivity with cells of the erythroid lineage at all stages of maturation in the peripheral blood, bone marrow, and fetal liver. Non-erythroid lineages are negative by flow cytometry. This MAb is useful in the diagnosis of erythroleukemia, identification of bone marrow erythroid precursors, gating erythroid nucleated precursor cells from malignant cells in bone marrow specimens.

Beskrivning: This antibody recognizes CD16 (FcγRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50-80 kDa. Two similar genes represent CD16, CD16A (FcγRIIIA), which exists as a hetero-oligomeric polypeptide-anchored form in macrophages and NK cells and CD16B (FcγRIIIB), which exist as a monomeric GPI-anchored form in neutrophils. Furthermore, there are two known polymorphisms of CD16B, NA-1 and NA-2. Individuals homozygous for NA-2 show a lower phagocytic capacity compared with NA-1. CD16 binds IgG in the form of immune complexes and shows preferential binding of IgG1 and IgG3 isotypes and minimal binding of IgG2 and IgG4. Upon IgG binding, both CD16 isoforms initiate signal transduction cascades that lead to a variety of responses including antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, degranulation and proliferation.

Beskrivning: Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.
